Abstract

The aim of this study was to evaluate the total polyphenol and flavonoid contents of Tagetes erecta L. (yellow and orange blossoms) petals extracts by using Folin-Ciocalteu assay and aluminium chloride colorimetric method respectively. Antioxidant activity, α-glucosidase inhibiting activity and the cytotoxicity were also evaluated in this study. The results revealed that the total polyphenol contents (150.18 ± 1.24 mg GAE/g DW), antioxidant activity (assessed by using DPPH method) and α-glucosidase inhibiting activity of T. erecta (yellow blossoms) petals in 70% ethanol were higher than those in the other three extracts including T. erecta (yellow blossoms) petals in 96% ethanol, T. erecta (orange blossoms) petals in 70% ethanol and 96% ethanol. There was a possitive correlation between α-glucosidase inhibiting capacity and flavonoid and polyphenol contents. These extracts of T. erecta in 500 µg/mL concentration exhibited cytotoxic activity against two human cancer cell lines, MCF-7 breast cancer cell lines and HeLa cervical cancer cell lines estimated by using sulforhodamin B assay. The greater amount of total polyphenol compounds leads to more powerful α-glucosidase inhibiting activity of T. erecta petals extracts.

Tóm tắt

Mục tiêu của nghiên cứu nhằm khảo sát hàm lượng polyphenol toàn phần (bằng phương pháp Folin-Ciocalteu) và hàm lượng flavonoid (bằng phương pháp so màu AlCl3) của các cao chiết từ cánh hoa vạn thọ thuộc 2 giống vạn thọ hoa vàng và vạn thọ hoa cam (Tagetes erecta L.). Hoạt tính kháng oxy hóa, ức chế α-glucosidase và gây độc tế bào cũng được khảo sát trong nghiên cứu này. Kết quả cho thấy, hàm lượng polyphenol toàn phần trong mẫu cao chiết bằng ethanol 70% của cánh hoa vạn thọ hoa vàng là 150,18 ± 1,24 (mg GAE/ g dược liệu khô) cũng như hoạt tính kháng oxy hóa (khảo sát bằng phương pháp khử gốc tự do DPPH) và hoạt tính ức chế α-glucosidase của mẫu cao chiết này cao hơn các mẫu cao chiết còn lại gồm cánh hoa vạn thọ hoa vàng dung môi ethanol 96%, cánh hoa vạn thọ hoa cam dung môi ethanol 70% và ethanol 96%. Kết quả thống kê cũng chỉ ra được có sự tương quan giữa hàm lượng polyphenol, flavonoid và hoạt tính ức chế α-glucosidase trong các mẫu cao chiết. Ở nồng độ 500 µg/mL, các mẫu cao chiết đều thể hiện hoạt tính gây độc tế bào trên 2 dòng tế bào ung thư vú MCF-7 và ung thư cổ tử cung HeLa (thực hiện bằng phương pháp Sulforhodamin B). Hàm lượng polyphenol và flavonoid trong mẫu cao chiết từ cánh hoa vạn thọ càng cao, hoạt tính ức chế α-glucosidase của mẫu cũng sẽ cao tương tự.
