Tuyển chọn và định danh dòng nấm hòa tan lân chịu mặn từ đất vùng rễ cây lúa hoang (Oryza rufipogon)
Abstract
Phosphorus is an essential element involved in many physiological and biochemical processes in plants; however, most phosphorus in soil exists in insoluble forms. Thirty-one phosphate-solubilizing fungal strains were isolated on NBRIP medium from the rhizosphere soil of wild rice (Oryza rufipogon) in Can Tho City in this study. Qualitative evaluation of phosphate-solubilizing ability showed PSI values ranging from 1.04 to 1.93. Based on quantitative analysis of soluble phosphorus, fungal strain OM9.2 exhibited the highest phosphate-solubilizing capacity, reaching 1,041.36 mg/L on day 10. Molecular identification using the ITS region revealed that strain OM9.2 showed the highest similarity to the Talaromyces genus in the NCBI database. In addition, strain OM9.2 demonstrated salt tolerance up to 6‰ NaCl. Notably, at 2‰ NaCl, this strain achieved the highest phosphate solubilization, exceeding 1300 mg/L on day 15.

Lân là nguyên tố thiết yếu tham gia vào nhiều quá trình sinh lý và sinh hóa của cây trồng, nhưng phần lớn lân trong đất tồn tại ở dạng cố định. Kết quả nghiên cứu này đã phân lập được 31 dòng nấm hòa tan lân từ đất vùng rễ cây lúa hoang (Oryza rufipogon) tại thành phố Cần Thơ. Định tính khả năng hòa tan lân với chỉ số PSI dao động từ 1,04 đến 1,93. Qua kết quả định lượng lân hòa tan, dòng nấm OM9.2 thể hiện khả năng hòa tan lân mạnh nhất, đạt 1.041,36 mg/L sau ngày thứ 10. Việc định danh phân tử vùng ITS cho thấy dòng nấm OM9.2 có mối quan hệ gần nhất với chi Talaromyces trên cơ sở dữ liệu NCBI. Bên cạnh đó, dòng nấm OM9.2 có khả năng chịu mặn đến nồng độ muối NaCl 6‰. Đồng thời, khả năng hòa tan lân của dòng nấm đạt giá trị cao nhất ở nồng độ muối NaCl 2‰, với hàm lượng lân hòa tan trên 1.300 mg/L sau 15 ngày nuôi cấy.
