Abstract
This study compared the effectiveness of two semen extenders: Beltsville Thawing Solution (BTS) and a modified BTS in which sodium bicarbonate was replaced with HEPES buffer (BTSH) during storage at 17°C for 96 h. The evaluated parameters included pH, osmotic pressure, sperm viability, membrane integrity assessed by the HOST, and sperm motility characteristics analyzed using a computer-assisted sperm analysis (CASA) system. The results demonstrated that BTSH maintained more stable pH and osmotic pressure, while significantly improving sperm motility and viability compared with BTS (p < 0.05), thereby reducing unfavorable fluctuations in the semen extenders throughout the storage period. After 96 hours of storage, BTSH achieved a viability of 73.67 ± 0.88% and total motility of 76.01 ± 1.79%, which were markedly higher than those obtained with BTS. Overall, the findings indicate that BTSH is the most suitable extender for boar semen storage at 17°C for up to 96 hours, effectively preserving overall sperm quality, motility, and plasma membrane stability throughout the storage duration.
