Abstract

Salt stress affects the growth, yield and quality of soybean. Mutagenesis through irritation method with gamma ray in combination with in vitro selection on the medium supplemented with NaCl was carried out on the soybean cultivar MTD 760-4 to obtain salt-tolerant callus lines. Results showed that, in salt concentration of 2.5 g/L, most callus grew normally. Salt tolerance to 5 g/L was achieved by culturing non-irradiated and irradiated callus at doses from 5-40 Gy on selection medium with 5 g/L of NaCl  after four times of selection. Callus tolerated salinity to dose of 7.5 g/L when selected at irradiated dose of 5 Gy. Proline content was accumulated highly in salt tolerant callus lines on media containing NaCl concentrations of  5 and 7.5 g/L.

Stress mặn ảnh hưởng đến sự sinh trưởng, năng suất và phẩm chất của đậu nành. Sự tạo đột biến bằng phương pháp chiếu xạ tia gamma kết hợp với chọn lọc in vitro trên môi trường nuôi cấy có bổ sung muối NaCl đã được thực hiện trên mô sẹo giống đậu nành MTĐ 760-4 nhằm tạo nên các dòng mô sẹo có khả năng chống chịu mặn. Kết quả cho thấy, ở nồng độ mặn 2,5 g/L mô sẹo đậu nành sinh trưởng bình thường. Sự chống chịu mặn 5 g/L đạt được khi nuôi cấy mô sẹo không chiếu xạ và mô sẹo được chiếu xạ với liều từ 5-40 Gy trên môi trường chọn lọc với muối NaCl 5 g/L sau 4 lần chọn lọc. Mô sẹo chống chịu mặn đến 7,5 g/L khi được chọn lọc ở liều chiếu xạ 5 Gy. Hàm lượng proline tích lũy cao trong các dòng mô sẹo chịu mặn ở môi trường chứa muối 5 và 7,5 g/L.
